Under Spanish law, Paul Dickov, Keith Gillespie and Frank Sinclair, who deny the charges, could stay in custody for up to two years before a trial is set.
The club and local MP Keith Vaz are pushing for the players to be brought home while the investigation continues.
The men were charged after three women claimed they were attacked in a hotel.
They could face up to 14 years in prison if found guilty.
The claims are the latest sex allegations against Premiership footballers in recent months.
Chief executive of the Professional Footballers Association, Gordon Taylor, said the media had a tendency to "blow up" such allegations because of the "pop star" status of players.
He said: "At the moment the three who are in prison are waiting for all the evidence to be checked out, and we would hope to know more by Monday as to whether it will be taken further or whether they will be released.
"Club officials and lawyers are out there and I know that some of the families of the three individuals were planning to go there.
'Unwritten law'
"All the players vehemently deny the charges against them.
"This was a birthday party and sometimes tours and things like that can get out of line, and then action will be taken.
"There will be a due process of law and if anybody is found to be acting improperly we have a strict disciplinary code.
"However we are disappointed when there is this assumption of guilt."
The men were moved on Saturday from a reception area of Sangonera jail near Murcia - dubbed the 'Fridge' - to cells reserved for alleged sex offenders.
"It is not a very good place to be. It is a big difference from the Hyatt Hotel where they were," he said.
